SyllabusStarting with Planar chromatography and moving to HPLC. Basic terms such as Tr ,K’, N and Rs are explained. The differences between normal and reverse phase and look at the common solvents used in HPLC and why they are used is explained. We explore the reasons why peaks have their characteristic shapes. How the building blocks of an HPLC fit together and how each component operates, looking at the various designs and their benefits. What are the critical areas that affect performance and how to maintain them. We chart the changes in column technologies and what the major advances have been. Interpreting chromatograms is discussed. Quantitation using external and internal standardisation. Integration using PC’s highlighting the many options that modern data systems allow users to adjust The mechanisms for separation are introduced with a look at the different functional groups that can be attached. Looking at reverse phase (C18, Phenyl and cyano columns) we show how pH affects separations and how effective HPLC is with different types of compounds. The course is punctuated with practical sessions to allow attendees to gain an enhanced practical feel of HPLC and to consolidate discussions and presentations. Attendees will be shown very basic method development with a quantitation practical to bring the training to a close |
